Abstract

Immature cotyledons of Rauwolfia serpentina were cultured on MS medium containing different concentrations and combinations of growth regulators. Analysis of variance exhibited highly significant differences among culture media combinations for callus initiation, formation of morphogenic calli and plantlet regeneration. Results showed that the growth hormone combination of NAA and BA was the most responsive during all culture phases and plant regeneration from cotyledon culture of Rauwolfia serpentina. A combination of 75% RH and 20�C exhibited the most optimal condition for survival of regenerated plantlets during hardening.
